When I looked at the data this graph shows I see that both the Wild type and also the yeast strain YPL068C have about the same linear pathway and so I came to the conclusion that even though the yeast strain YPL068C has somewhat a deviation difference but its still small enough and somewhat close enough to the wild types and so therefore both of them are yet resilient to the pH 4 stress test.

YPL068C is neither more or less sensitive to the cold than the wild type strain. This is shown because the doubling times change the same amount before and after the cold shock in both the wild type strain and YPL068C.

In a BY4735 background, knocking out YPL068C seems to have a moderate effect on growth rate in log-phase. In this assay, the BY4735 strain's doubling time was 124 minutes, while the YPL068C knock-out strain's doubling time was 209 minutes.

Ultraviolet Sensitivity

YPL068C is more sensitive than BY4735(wild) given that there is less colonizes present when under the same stress.

G-418 Stress

image008.png

In the BY4735 background, knocking out YPL068C seems to have a little effect on the growth rate. In the knock-out experiment, the BY4735 strain's doubling time was 149 minutes, whereas the YPL068C knock-out strain's doubling time was 235 minutes. The calibration experiment, the BY4735 strain's doubling time was 64 minutes, whereas the YPL068C knock-out strain's doubling time was 92 minutes.

Heat Shock

HeatShockforYP.png

Heat shock had no change on the doubling time, but the starting amount of yeast cells less. The heat seemed to kill off the cells, the longer they were in the hot water bath.

Methanol Sensitivity

The wild type had a doubling time of 150 min, the YPL068c had a doubling time of 200 min. The doubling time increased which means this strain is sensitive to methanol.

Cold Shock Sensitivity

Protocol: Cold Shock

YPL068C is neither more or less sensitive to the cold than the wild type strain. This is shown because the doubling times change the same amount before and after the cold shock in both the wild type strain and YPL068C.
