UW-Stout/Heat Shock FA21
YHLO12W (B)
- When stressing out this strain of yeast, it had no growth. On the control plate, there was normal growth. So we can see that this yeast strain is sensitive to heat.
Nitrogen Starvation
- There was no significant change between the plates with NH₄ and without. They seemed to be growing at the same pace. So this particular gene was not affected by the Nitrogen Starvation stress.
UW-Stout/Bud Scars FA21
Cells grown with this gene (YHL012W) knocked out had an average of 1.20 bud scars per cell while the wild type (BY-RY4735) had 0.85 bud scars per cell. Of 106 YHL012W cells counted, 127 bud scars were seen. This indicated that the YHL012W gene slows reproduction rate and thus bud scarring in yeast cells.

- The Yeast Strain of YHL012W was the one that was the most affected by the 18.5 % Ethanol it still has a steady growth curve

The YHL012W yeast strain doubling time without any stress was 187 minutes which is very similar to the wild type with 182 minutes. When both strains were stressed by G418 they YHL012W doubling time was 209 minutes and the wild type was 178 minutes. (These times are averaged between three trials)

UW-Stout/Copper FA21
Looking at this graph you can see that something quite odd is happening. At first, we thought this was some technical error of some sort, but we ran the experiment again and got the exact same result. Looking at this data and how we explain this oddity below, we believe that we cannot properly calculate the average doubling time for this strain of yeast while stressed. The unstressed version's average doubling time is about 377 minutes.
- In these images you can see the growth of the yeast. In the right image this is a normal growth of yeast and in the left one we can see the yeast clumped up in the center of the yeast cell. This is strange because the image in the left is showing the yeast floating on top of the Copper Sulfate solution and it is collected in the center of the well which is different from the normal yeast which is evenly distributed in the well. The yeast that was different was the YHL012W strain, so this indicates that the yeast reacted differently versus the others. The reason our data looks the way it does is because we believe the way the machine determines the size of the growth is getting thrown off because of the massive concentration in the center. Why is this happening? We are not sure and further experimentation is required to get a definite answer.
