Protein Function/Process

Diacylglycerol pyrophosphate (DGPP)phosphatase, product of DPP1, purified to homogeneity. Pure enzyme utilizes DGPP and phosphatidic acid (PA) as substrates. DGPP is the preferred substrate with a 10-fold higher specificity constant. [5] [9]


DPP1-encoded diacylglycerol pyrophosphate (DGPP) phosphatase is induced by inositol supplementation and in stationary phase. DGPP phosphatase activity is inhibited by the phospholipid intermediate CDP-diacylglycerol. The DGPP molecule regulates phospholipid synthesis by activating phosphatidylserine synthase [7] [9]


The 3-domain lipid phosphatase motif in diacylglycerol pyrophosphate (DGPP) phosphatase is essential for catalytic activity using both DGPP and phosphatidic acid as substrates [4] [9]


DPP1 was identified based on the protein sequence of diacylglycerol pyrophosphate (DGPP) phosphatase. Accounts for nearly all of the DGPP phosphatase activity in the cell. Remaining DGPP phosphatase activity is due to the LPP1-encoded enzyme. DPP1 gene product also exhibits phosphatidic acid phosphatase activity, but with a 10-fold lower specificity. [3] [9]

The DPP1-encoded diacylglycerol pyrophosphate (DGPP) phosphatase is localized to the vacuolar membrane. [2] [9]
