The Kupiec lab is looking for highly motivated and independent researchers to join our group in exciting projects investigating the mechanisms that create mutations, and those that repair DNA and prevent genome instability. Previous experience working with yeast is considered an advantage, but not essential.
A post-doc position is open in the laboratory of John McCusker (https://mgm.duke.edu/faculty-and-research/primary-faculty/john-h-mccusker-phd/) in the Department of Molecular Genetics & Microbiology at Duke University Medical Center on a newly funded R01 to work on the quantitative genetics of drug resistance in Saccharomyces cerevisiae.  The post-doc position requires demonstrated productivity (i.e. publications) as well as expertise in genetics (preferably microbial, ideally with S. cerevisiae) and in molecular biology. Enzyme reactions have long been analyzed in vitro, using pure enzymes and diluted buffer conditions. Due to the large amount of data generated and collected with thousands of enzymes, enzymology has made tremendous progress on understanding the incredible power of biocatalysts. However, dilute, in vitro conditions are far from the surroundings of natural enzymatic reactions that take place inside cells. The cellular medium is more accurately described as a heterogeneous crowded gel, dense and filled with all sorts of macromolecules and cellular lipidic organelles which may result in some partitioning effects and changes in diffusion. Therefore, enzymatic parameters determined using classical enzymology setups may not perfectly represent the real, in vivo based, rate and equilibrium constants. Although some advances have been made toward the comprehension of viscosity and crowding effects, we are still far to derive rate and equilibrium parameters from in vivo enzymatic reactions.

The project ENZINVIVO will address this issue focusing on two isoforms of phytoene synthase (carotenoid biosynthesis) as model enzymes. The enzymatic properties of these enzymes will be investigated in vitro, through a set of measurement in environments of increasing complexity and in vivo expressing the carotenoid biosynthesis pathway in S. cerevisiae and using synthetic biology tools and concepts to tune, at will, substrate and enzyme levels.

- how the intracellular medium influences enzymatic reactions.
 
- how can we build an in vivo approach to measure enzymatic parameters.
 
- how general models of enzymatic equations can be rewired to account for the complexity in vivo approaches. Our work will notably verify (or not) whether a Michaelis-Menten description of the kinetics remains valid in vivo.
 
The candidate will be first responsible for the construction of engineered strains with fine-tuned or inducible expression of CtrE (geranylgeranyl pyrophosphate synthase, GGPP synthase) to modulate intracellular concentration of GGPP, substrate of the phytoene synthase in a range consistent with its KM. Then, he/she will be in charge of analyzing the consequences of the heterologous pathway on the yeast physiology through multi-levels characterization of the recombinant strains.

Post-doctoral position on comparative transcriptomics in S. cerevisiae strains (posted 9th June 2022)

Mar Albà, ICREA Research Professor, Evolutionary Genomics Group, Hospital del Mar Medical Research Institute (IMIM) – Barcelona Biomedical Research Park (PRBB), Barcelona.

Description: Looking to recruit one highly motivated researcher at the postdoctoral level to perform research on comparative genomics, transcriptomics and translatomics with the goal to better understand the molecular basis of the evolution of new genes in yeast. The position is funded by an ERC Advanced Grant (NovoGenePop) and will have an initial duration of three years. The researcher will study transcriptome diversity in wild and domesticated populations of yeast. She/he will develop computational approaches to analyze a large amount of RNA-seq and Ribo-seq data from different S. cerevisiae strains and Saccharomyces species, characterize the transcriptomic diversity in different populations and develop methods to identify recently evolved genes. Mutations and selective signatures associated with de novo gene birth will be identified using genomics data from a large number of strains.

Requirements:

  • Research experience in comparative genomics and transcriptomics
  • Expertise in at least one programming language
  • Expertise in a programmatic statistics language (ideally R)
  • Sound knowledge of population genetics and molecular evolution

Description: The Truman laboratory in the Department of Biological Sciences at UNC Charlotte employs a range of molecular technologies including mass spectrometry, CRISPR-genome engineering and high-throughput screening to study the role of post-translational modifications on Hsp70 (see: http://www.trumanlab.org). We are seeking a highly motivated postdoctoral researcher with experience working with budding yeast.

Ideal Qualifications:

  • Ph. D in biochemistry, molecular biology, cancer biology or related field.
